Initiator Caspases-8 and -12 in the Processes of Realizing Selected Stages of Human Lymphocyte Apoptosis by UV-Radiation

Abstract

Based on the analysis of the changes in the level of the functional activity of human lymphocyte initiator caspases-8 and -12 after exposure to UV radiation (240–390 nm) at a dose of 1510 J/m2, we have shown the possibility of their participation in the realization of the receptor caspase pathway (caspase-8) and signaling pathways associated with disruption of calcium homeostasis and changes in the level of secondary messengers such as Ca2+ and cAMP (caspase-12). The flow cytometry method has shown that 4 hours after UV radiation of lymphocytes, the number of cells in the early stage of apoptosis increased in comparison with the control by 20%. This data can be explained by the activation of «fast-acting» apoptosis signaling pathways leading to translocation of phosphatidylserine to the cell surface.
