Abstract
The catalytic activity of free collagenase from Clostridium histolyticum under the influence of UV light at the doses of 151–6040 J/m2 was modulated to a greater extent than in the immobilized on the chitosan matrix condition. After UV irradiation of the enzyme solution with a dose of 3020 J/m2, its activity decreased by ~9%. After immobilization of collagenase on the matrix of acid-soluble medium-molecular and high-mole-cular chitosans, no change in the enzyme activity was detected in the entire range of the doses used by the authors. Absorption on chitosan leads to an increase in the stability of heterogeneous biocatalysts with respect to UV irradiation in comparison with the free form of bacterial collagenase. The formation of bonds between the enzyme and matrix of both medium-molecular and high-molecular chitosans has a significant effect on the stability of the immobilized preparation towards UV radiation, probably due to an increase in the rigidity of the tertiary structure of collagenase during adsorption.
