Estimation of the Effects of Radiation Countermeasure Agentson the Expression of Mouse Bone Marrow Genes and microRNAsin a Remote Period after Low Dose Rate γ-Radiation

Abstract

F1 (CBA × C57Bl) mice were exposed to prolonged (21 h) γ-radiation with a low (10 mGy/min) dose rate at the large dose (12.6 Gy) in standard housing conditions (5 control and 5 under a preparation usage). The following group variants were studied: betaleukine (rh interleukine 1β) 50 mkg/kg 2 h before irradiation or 3 mkg/kg 22 h before irradiation or riboxin 300 mg/kg 0.5 h prior exposure. In control groups, we revealed increasing P53 and miR-145 expression but decreasing miR-125 expression 3–4 months after exposure. It was shown that in the protected groups betaleukine-50 and riboxin significantly increased P53 expression in relation to the exposed control during the period of 3–6 months. In the same time, betaleukine-3 insignificantly increased P53 expression or even decreased its expression. Betaleukine in both doses increased miR-145 expression and decreased miR-125 expression. It has been observed that for betaleukine there is a connection between P53 overexpression in a remote period and its decrease prior to exposure. It allows us to propose that P53 expression in a remote period can play a role of the genom instability marker.
